Resveratrol inhibits lung cancer by targeting cancer stem cells and regulating the cancer tumor environment.
Abstract
Increasing evidence indicates that cancer stem cells (CSCs) are key drivers of the initiation and recurrence of malignant tumors. Cellular components of the tumor microenvironment (TME) affect cancer initiation and progression, such as cytokines and chemokines, therefore targeting CSCs and TME is a novel anti-cancer approach. Resveratrol (RES), a bioactive phytochemical extracted from various plants, Demonstrated tumor suppressive activity in lung cancer. But the mechanism remains poorly understood. Our data show that the expression level of IL-6 is positively associated with the presence of lung cancer-like stem cells (LCSC) in human lung cancer tissue. In vitro results showed that IL-6 was significantly elevated in spheroid-forming lung cancer cells. and can increase the stability of LCSC cells and their ability to form tumor spheres. The percentage of CD133-positive cells and the expression of LCSC-specific markers (CD133, ALDH1A1, and Nanog) at the same time. Our experimental results confirmed that Resveratrol effectively inhibits the properties of LCSCs. Reduce the transmission level Wnt/β-catenin and reduced IL-6 levels both in vitro and in vivo. We also found that treatment with resveratrol causing stimulation of signal transmission Wnt/β-catenin depletion by LiCl (GSK3β activator) RES also reversed the properties of LCSCs promoted by IL-6 and signaling. Wnt/β-catenin When taken together These data show that Resveratrol inhibits lung cancer by targeting LCSCs and IL-6 in the TME. This new study provides evidence that Resveratrol shows multi-target effects in inhibiting lung cancer and is a potential cancer prevention compound.
