Resveratrol has its antioxidant and anti-inflammatory protective mechanisms decreased in aging
In the elderly, the effects of oxidation and inflammation are increased. Resveratrol (RSV) is a polyphenol with many proven biological activities. For example, it is an antioxidant and anti-inflammatory. Therefore, the aim of our study was to investigate the antioxidant and anti-inflammatory effects of RSV in human mononuclear cells (PBMCs) from people aged 40 to 59 and 60–80 years. Six to eight patients were selected according to age group. Cells were isolated and divided into 4 groups: control group (C), RSV only, H2O2 (to induce an oxidizing environment – C+) and H2O2+RSV. Quantification of reactive nitrogen species (NO and ONOO-) as well as pro- and anti-inflammatory cytokines (TNFα, IL-6 and IL-10) was performed. Pearson correlation and between-group comparisons (p<0.05) were performed. Our results showed that RSV was more active in the middle-aged group compared to the elderly group. This is related to the NO/ONOO- balance and the levels of the cytokines IL-6 and TNFα. It is also possible to observe an improvement in the anti-inflammatory profile in both age groups. But it was more effective in cells in the middle-aged group. Therefore, we observed that RSV had better effects in reducing key biomarkers of oxidation and inflammation.
